When assigning a flow to a trunk, there are two options available. If it's your first time publishing the Flow (option 1), you can complete the quick wizard to assign it to your trunk. Alternatively, if the Flow is already published (option 2) or you wish to add it to another trunk group, you can do so from the Flow Builder tab. Both options are linked in the table of contents below.

Inbound Trunk Group(s) are used to handle inbound calls. Most accounts will only have one trunk group, if you have more, then you need to select the trunk you want the Flow associated with.
Route Profile(s) are used to handle routing for DIDs. Here you will want to create a new profile unless you have already created one previously that you want to associate this flow with.
Route Profile Route(s) are used to determine where the DID should be routed to. Each Route Profile Route can be assigned a priority, destination type, and destination. The Route Profile Route is used to determine which route is selected when an incoming call is handled
Voice API Profile(s) are used to determine how inbound calls are handled. Similar to your trunk selection, pick the service trunk and outbound trunk this Flow will be associated with.
Review to make sure all the settings look good and submit!
